Chairman Trevor Bull hopes the present day Seadogs can follow in the footsteps of the former Scarborough FC club and one of their National North rivals:
"I went to Wembley for the final last year because, of course, Spennymoor, were in it from our league. So if Spennymoor can get to it, who knows? But we're not putting any pressure on anybody to achieve anything. We're just going to go out there and do the best we can and see where it takes us, but it'd be great to have some fun in the Trophy given our former club's rich history in that competition."
The winners of the tie receive £3750, the losers receive £1000.
The Seadogs come into the game after back-to-back victories, over Curzon Ashton and Guisborough Town.
Boro won't be taking Leek lightly after going out of the FA Cup at another NPL Premier side, Ashton United.
